On the basis of optical stiffness, an optoelectronic device with nanometer resolution is able to monitor the offset displacement of cell movement when it is subjected to the forces on the order of picoNewton strength.
The structured beams resulted from holographic reconstruction improve the precise position of multiple cells, and service the force measurement on the order of picoNewton, which have considerable potential for exploration in the molecular mechanism of cellular elasticity, adhesion, and migration.
The isolated cell attributed to optical tweezers could be extracted by pipet and placed in the circular pool, so that Raman spectroscopy technique explored the heterogeneity among the four cells. Stable isotope labellings of microbial cells with specific functions have been the subject of metagenomics in the past decade.
In the experiment, the optical tweezers have excellently undertaken the precision relocation of bacterial particles for the measurement on Raman spectrum. In Raman-activated live cell sorting, the throughput of 500 cells per hour and the accuracy of 98.3 ± 1.7% would be widely applied in the downstream DNA analysis or phenotypic definition.
Due to these properties, optical trapping has become a key technology for biological experimentation since it enables precise control over the position of and applied force on objects at the micron and submicron scale.
To activate the variety of photosensory domains developed for cellular signaling pathway control, there must be complementary development of optical tools for cell culture illumination. However, the current optical methods lack practical applicability to routine cell culture stimulation and/or lack critical characterization and functionality.
